N-terminal truncated RHT-1 proteins generated by translational reinitiation cause semi-dwarfing of wheat Green Revolution alleles

چکیده
The unprecedented wheat yield increases during the Green Revolution were achieved through introduction of Reduced height (Rht)-B1b and Rht-D1b semi-dwarfing alleles. These Rht-1 alleles encode growth-repressing DELLA genes containing a stop codon within their open reading frame that confers gibberellin (GA)-insensitive semi-dwarfism. In this study, we successfully took hurdle detecting wild-type RHT-1 proteins in different organs confirmed degradation response to GAs. We further demonstrated Rht-B1b produce N-terminal truncated translational reinitiation. Expression these transgenic lines Rht-D1c, an allele multiple copies, ability cause strong dwarfism, resulting from insensitivity GA-mediated degradation. detected spikes nodes, but not aleurone layers. Since dwarfism have dormancy, finding suggests tissue-specific differences reinitiation may explain why reduce plant without affecting dormancy. Taken together, our findings only reveal molecular mechanism underlying also demonstrate main occurs plants.

Molecular characterization of Rht-1 dwarfing genes in hexaploid wheat.
The introduction of the Reduced height (Rht)-B1b and Rht-D1b semidwarfing genes led to impressive increases in wheat (Triticum aestivum) yields during the Green Revolution. The reduction in stem elongation in varieties containing these alleles is caused by a limited response to the phytohormone gibberellin (GA), resulting in improved resistance to stem lodging and yield benefits through an incr...

The effects of dwarfing alleles (reduced height, Rht) in near isogenic lines on wheat grain quality are characterised in field experiments and related to effects on crop height, grain yield and GA-sensitivity. Alleles included those that conferred GA-insensitivity (Rht-B1b, Rht-B1c, Rht-D1b, Rht-D1c) as well as those that retained GA-sensitivity (rht(tall), Rht8, Rht8 + Ppd-D1a, Rht12). Full ch...

Wheat is a major staple food crop providing about 20% of dietary energy and proteins, and food products made of whole grain wheat are a major source of micronutrients like Zinc (Zn), Iron (Fe), Manganese (Mn), Magnesium (Mg), Vitamin B and E. Wheat provides about 40% intake of essential micronutrients by humans in the developing countries relying on wheat based diets. Varieties with genetically...
